AUFGABENBESCHREIBUNG:
  • Define and develop Next Generation Sequencing (NGS) analysis pipelines for quantifying CRISPR based on-target and off-target events as well as algorithms for computational prediction of guide RNA specificity and efficiency
  • Routinely, analyze and interpret NGS sequencing data to determine gene editing efficiencies and off-target events
  • Work closely with colleagues in the experimental team performing CRISPR experiments to communicate results or advise on appropriate bioinformatic analyses
  • Troubleshoot and optimize, both independently and in collaboration with colleagues
  • Participate in regulatory interaction meetings, provide responses to questions from regulators, and strategically communicate off-target characterization results

ERFORDERLICHES PROFIL:
  • Degree (Ph.D., or M.S. with 6+ years of relevant experience) in computational biology/bioinformatics or related field
  • Background in molecular biology, particularly gene editing with relevant experience in academic or industrial setting
  • Experience in developing computational pipelines to analyze NGS datasets, including genome/exome sequencing data and/or targeted DNA sequencing
  • Experience in developing and maintaining pipelines for CRISPR-Cas analysis (e.g. GUIDE-Seq, SITE-Seq, Amp-Seq)
  • Confidence in use of common bioinformatics tools for NGS data analysis (e.g. BWA, BEDtools, GATK, IGV) and public omics databases (e.g. gnomAD, TCGA)
  • Proficiency with Python and/or R is essential; familiarity with other languages such as Perl, C++ and MATLAB would be a plus
  • Strong understanding of CRISPR-Cas technology

Nice to Have:

  • Experience with designing guide RNA and in silico assessment of on/off-target activity
  • Familiarity with Illumina sequencing platforms and NGS library preparation methods
  • Experience in analysis of single cell sequencing data (e.g. single cell DNA sequencing, MissionBio)
  • Proficiency with handling large-scale genomics data in a Linux environment
  • Experience with front-end web development (e.g. HTML/CSS/Javascript, bootstrap and/or R Shiny) is considered a plus
  • Strong communications skills (written and oral)
  • Attention to detail and problem-solving skills, strong communication skills, and the ability to work in a team
